摘要
应用奶牛乳房炎大肠杆菌分离株灭活菌苗免疫健康家兔,制备兔抗大肠杆菌高免血清。对该大肠杆菌菌体裂解产物进行SDS-PAGE并进行Westernblot分析,结果该菌株与稀释一定倍数的兔高免血清反应,产生3条特异性带,其中约33ku蛋白为主要免疫原性蛋白。对其进行电洗脱纯化,得到单一条带的目的蛋白。
In order to prepare antiserum against Escherichia coli of rabbits, healthy rabbits were immunized by antiserum against Escherichia coli isolated from dairy cows suffering from mastitis. Lysate of the isolated E.coli was analyzed with SDS-PAGE and Western blot. The results showed that the lysate could react with diluted antiserum to produce three bands and the band of main immunogenic protein was at 33 ku. The main immunogenic protein could be purified to show a single band by electroneluting.
