摘要
目的对黑水缬草愈伤组织诱导培养基和生长的培养条件进行研究,构建黑水缬草快速繁殖体系。方法通过筛选外植体,添加不同种类及浓度的生长调节剂对黑水缬草进行愈伤组织诱导、芽增殖培养及生根培养。结果诱导愈伤组织适宜培养基:MS+0.5mg/LNAA+5.0mg/L6-BA。芽增殖培养以MS+0.2mg/LNAA+10.0mg/L6-BA为宜。生根培养以MS+0.1mg/LNAA为佳。结论利用组织培养技术能够实现黑水缬草快速繁殖。
Objective To provide some new evidences for rapid propagation of Valeriana amurensis Stair. ex Kom through studying on medium with callus tissue culture and the condition of growth. Methods Primary culture of different explants, culture of cluster buds and their rooting culture were conducted on medium of treatment combinations of adding different hormones. Results The appropriate medium for different culture stages were MS ＋ 0.5 mg/LNAA ＋ 5.0 mg/IA-BA in callus culture induction, MS ＋ 0.2 mg/LNAA ＋ 10.0 mg/L6-BA in differentiation and subculture of cluster buds, MS ＋ 0.1 mg/LNAA in rooting culture. Conelusion Rapid propagation can be achieved with plant tissue culture in Valeriana amurensis Stair. ex Kom.
