2006年吉林省野生型麻疹病毒分离鉴定及核蛋白基因序列分析

摘要 目的:研究吉林省麻疹病毒的基因型别及核蛋白(N)基因序列特征,探讨控制麻疹发生的措施。方法:采用B95 a和Vero/SLAM细胞分离培养麻疹病毒,通过逆转录-聚合酶链反应(RT-PCR)方法从分离的麻疹病毒中扩增核蛋白(N)基因羧基端450个核苷酸(bp)片段,进行核苷酸测序分析并与基因库GenBank中麻疹病毒的各代表株基因序列的同源性比较。结果:分离到8株麻疹病毒,N基因与H1基因型代表株[1]相比同源性为98.8%。结论:吉林省麻疹流行株属于H1基因型。 Objective:To study the genetic characteristics and genotype of measles strains and take measures to control measles in Jilin Province. Methods:Throat swabs were collected for virus isolation with B95a and Vero/SLAM cells. A 450 nucleotides fragment of the C - terminal of the nucleoprotein ( N ) gene was amplified by RT - PCR and subjected to sequence and phylogenetic analysis using Bio -Edit software. Results:Eight strains were isolated, N genes homogeneity was 98. 8% compared with other wild -type viruses of H1 genotype. Conclusion:Jilin Province's circulating measles viruses belongs to clade H1.
