提取人血浆中β2GPI的一种简便高效方法及其在自身免疫病诊断中的应用研究 被引量：3

摘要 目的:利用聚乙二醇(PEG)4000沉淀配合亲和层析和离子交换层析的方法从人血浆中提取β2GPI蛋白,并研究相关的临床意义。方法:用180 g/L的PEG4000从人血浆沉淀含β2GPI的组分,所得组分重溶后进行Heparin-Sepha-rose CL-6B亲和柱层析和Source 15 S离子交换柱层析,用分离纯化的β2GPI和购买的纤溶酶原(Plasminogen,PLG)建立反复妊娠丢失患者血清抗体抗β2GPI和抗PLG抗性的ELISA检测方法。结果:经SDS-PAGE和Western blot检测,本方法分离提取得到的蛋白为纯度大于95%的β2GPI。抗β2GPI和抗PLG抗性的检测结果呈正相关关系(r=0.750),且两者检出的阳性率无统计学意义(P>0.05)。结论:结合了PEG沉淀和两步层析方法可以有效地从人血浆中提取得到β2GPI蛋白用于相关检测和研究。
