摘要
本文采用多聚酶链反应技术及单链构象多态分析(PCR/SSCP)检测了34例口腔鳞癌及20例癌旁组织中P53基因Ⅴ、Ⅷ外显子,发现癌组织中20例出现基因突变,阳性率为58.8%,而在癌旁组织中无1例出现突变,本研究表明,口腔鳞癌的产生与P53突变有关。
Exons Ⅴ Ⅷ of the P 53 gene in 34 cases of oral mucous squamous cell carcinoma and 22 cases of tissues immediately adjacent to carcinoma were studied using polymerase chaine reaction-single strand conformation polymorphism andysis (PCR-SSCP).The incidence of P 53 mutations was 58.8%(20/34)in oral mucous squamous cell carcinoma, and no mutation was discovered in all case of tissues immdiately adjacent to carcinomas. The mutations were more frequently found in the poor differentiative carcinoma than that in the higher differentiative ones, although no statistical signifience was noticed. On the other hand,we also analysed the corretation of the smoking history with the incidence of P 53 gene mutation,18 out of 24 smokers had P 53 gene mutation whereas 2 out of 10 non-smokers in contrast were found to have P 53 gene mutation (P=0.0184). The results suggest that the carcinogenesis of oral squamous cell carcinoma may be retated to the mutatons of P 53 gene and smoking may stimulate the mutation of P 53 gene, perhaps it play an important role in the carcinogenesis of oral mucous. gene
