纤维粘连蛋白酶切片段的制备及其对大白鼠血浆中冷凝蛋白形成的抑制作用

摘要 冷凝蛋白(coldinsolubleprotein,CIP)是机体血浆经低温致冷后形成的大分子冷不溶性蛋白,它在机体组织冷损伤起着一定的作用。CIP的形成与纤维粘连蛋白(fibronectin,Fn)有关。本文观察到猪血浆Fn的胰蛋白酶酶切片段混合物(Fnfragments,Fnf)在5×10-6mol/L浓度时,能明显抑制新鲜大白鼠血浆中CIP的形成,血浆中CIP的形成量(79±46μg/ml)仅为对照血浆组(781±260μg/ml)的10%,推测Fn分子上可能有相应的结构参与了CIP的形成。 CIP(cold insoluble protein) is a cold insoluble protein with large molecular weight.In the present study, fibronectin was separated and purified from fresh pig plasma with affinity chromatography, and fibronectin fragments(Fnf) mixture(Wm=60kD) was obtained by the method of fixed trypsin. While Fnf was added into fresh rat plasma up to 5×10 -6 mol/L, CIP formation(79±46μg/ml)was inhibited markedly, only 10%(15℃) as much as that of the control group(781±260μg/ml). The results suggested that some part of the structure(s) of Fn molecule might take part in the plasma CIP formation.
