摘要
用密闭克隆培养法观察了人舌鳞状细胞癌Tca8113细胞的热敏感性和热耐受性。结果显示:43℃和45℃水浴加热对Tea8113细胞有明显杀伤作用。第一次加热能诱导细胞对再次加热产生热耐受(Thermotolerance),加热后6h已能显示热耐受产生,并持续72h后消退。热耐受能降低热效应对细胞的杀伤作用,在临床肿瘤热疗中,重复加热必须避开热耐受的起动和持续期。
Heat Sensitivity,Kineties of thermotolerance induction and decay were studied in Human Tongue Squamous Cells (Tea8113) in vitro by from of colony forming rate. The Do of the heat Survival curves of Tea8113 Cells at 43℃, 45℃ were 21. 6min, 6. 9min respectively, 41℃ heating did not show any citotoxicity. The kinetics of thermotolerance induction were measured with a triggering dose of 43℃/20min, or 45℃/10min. The cells were then challenged with 43℃ or 45℃ after incubation for 6, 12, 24, or72h at 37℃. At 43℃ /20min trigging dose,thermotolerance was maximum at 6h (TTR=2. 4) and decayed completely at 72h At 45℃/10min priming heating,, themotolerance was maximum at 12h (TTR=3. 9) and decayed completely at 72h too. The present results have some implication for the choice of fractionation regiment in clinical treatment of cancer with hyperthermia.The intervals between two fraction hyperthermia must be longer than 72h to ensure the completet decay of thermotolerance.
