The Relationship Between Expression of Survivin mRNA and Protein with Apoptosis in Colorectal Carcinoma

摘要 目的研究Survivin基因 mRNA拷贝数、蛋白表达在大肠癌诊断、预后等方面的意义及与细胞凋亡间的关系。方法分别采用分子信标荧光绝对定量PCR、S-P免疫组化和TUNEL法,检测Survivin mRNA拷贝数、蛋白表达及细胞凋亡指数。结果Survivin mRNA拷贝数和蛋白表达阳性率,肿瘤组织显著高于正常组织;凋亡指数AI,肿瘤组织显著低于正常组织(P<0.05)。肿瘤组织中Survivin mRNA拷贝数与淋巴结转移和病理类型显著相关;蛋白表达阳性率与肿瘤部位和患者生存状况显著相关,蛋白表达阴性组AI显著高于阳性组(P<0.05)。Survivin mRNA拷贝数与蛋白表达无相关性(P>0.05)。结论Survivin mRNA和蛋白在大肠癌中表达上调,可作为判断预后的生物学指标。 Objective To study the significances of survivin mRNA and protein expression in colorectal carcinomas and the relationship between them with apoptosis index (AI). Methods The copies of survivin mRNA were measured by quantitative PCR with a molecular beacon probe. The expression of protein was investigated by immunohistochemistry SP method. AI was analyzed with TUNEL. Results The copies of survivin mRNA and the positive rate of protein in colorectal carcinoma were significantly higher than those in normal tissues. In contrast, AI in colorectal carcinoma was significantly lower than those in normal tissues ( P 〈 0.05 ). The absolute copies of survivin mRNA were related with lymph node metastases and different histological types. The positive rate of protein expression was related with tumor locus and survival. AI in negative cases of protein expression was significantly higher than in positive cases ( P 〈 0.05 ). There were not correlations between the copies of survivin mRNA and protein expression ( P 〉 0.05 ). Conclusion The copies of survivin mRNA and the positive rates of protein expression were higher in colorectal carcinoma. It may be a potential molecular indication to predict the prognosis.
