巴西虫草液体培养条件的优化 被引量:6

摘要 以菌丝体生物量为指标,采用正交实验设计方法对巴西虫草液体培养的摇瓶生长条件进行了优化研究。结果表明:优化培养基的组成为可溶性淀粉2%、蛋白胨1%、大豆粉1%、KH2PO40.1%、MgSO4.7H2O 0.15%、VB10.01%、FeSO4.7H2O 0.002%、CaC l20.1%。该培养基下的菌丝体生物量为19.11g.L-1,比原培养基的发酵得率提高了26.01%;对巴西虫草液体培养的外部条件进行优化,确定摇瓶培养条件为:培养基初始pH 6.5、装液量75mL/250mL、接种量7.5%、菌龄4d、发酵时间4d。研究了优化后培养条件的菌体生长代谢曲线,并得到了巴西虫草的液体培养条件和生长动力学曲线,为工业化生产提供了一定的理论依据。 The submerged culture conditions of Cordyceps brasiliensis Henn were optimized by orthogonal test. The results showed that the optimal medium ingredients were as follows.- soluble starch 2% , peptone 1% , soybean 1% , KH2PO4 0. 1% , MgSO4 .7H2O 0. 15% , VB1 0. 01%, FeSO4· 7H2O 0. 002%, and CaCl2 0. 1%. The mycelial biomass was 19. 11 g · L^-1 with this optimal medium to culture the fungus. The optimized external conditions were initial pH 6. 5, medium capacity 75mL/250mL, seed age 4d, inoculum size 7.5% and culture time 4d. The growth curve of C. brasiliensis under the optimized condition was also investigated, and the mycelium yield was 19. 11 g · L^-1 which was 26. 01% higher than with original medium.
