摘要
观察雌激素对卵巢癌细胞的增殖调节。利用卵巢上皮性癌AO、3AO细胞,采用增殖细胞核抗原免疫组化检测法及3H-胸腺嘧啶掺入率测定法观察了不同浓度的雌二醇(E2)对卵巢癌细胞增殖的影响。结果:两株细胞对10-5~1013 mol/L之间的17-β-E2具有相似的剂量反应。AO细胞在10-7~10-9mol/L浓度范围中增殖明显,而超出该范围这种作用由促进转向抑制; 3AO细胞在 10-9~ 10-13 mol/ L之间增殖明显。两者在10-5mol/ L的 E2的作用下均呈现明显的抑制作用、结论:雌激素对卵巢癌的增殖作用具有剂量依赖性,高浓度的雌激素对癌细胞有明显的抑制作用。
PURPOSE To evaluate the effects of 17-β-estradiol on cell growth. METHODS The effects of estradiol on the growth of human epithelial ovarian cancer cells lines AO, 3AO were studied by using of PcNA and [3H]-thymidine uptake. Cells were cultured under estrogen- deprived condition. RESULTS Estradiol regulated cell growth dose dependantly in the range of 10-13 - 10-5 mol / L in both cell lines. Estradiol stimulated AO cell proliferation at 10-9 - 10-7 mol / L but not at others, and 3AO cell lines between 10-13 mol / L and 10-9 mol / L but not at all lower concentrations. A greater inhibition of cell proliferation was produced by estradiol at 10-5 mol / L. CONCLUSIONS The proliferation of these two cells can be regulated dose-dependantly by estrogen. Cell growth can be inhibited in great degree by estrogen at high concentration.
