摘要 应用RT-PCR方法检测30例胃腺癌mdr-1基因的表达。结果发现术前化疗的胃腺癌中mdr-1表达阳性率为91%,明显高于术前非化疗组的阳性率(47%)(P<0.O1)。在术前非化疗组中,中分化胃癌的阳性率高于低分化胃癌的阳性率(P<0.05)。mdr-l基因的表达与胃癌转移的发生率无明确关系。临床检测胃癌mdr-l基因的表达,有助于选择术后化疗方案。 Resistance to anticancer agents is the major obstacle to controlling malignant tumors. To characterize multidrug resistance (MDR) phenotype in human gastric carcinoma,expressions of the mdr-1 gene in 30 cases of gastric carcinoma were assayed by polyrnerase chain reaction(PCR) of RNA after reverse transcription (RT). The expression level of the cases after chemotherapy was much higher than that of cases without chemotherapy (P<0.01). Moderately differentiated gastric carcinomas showed a higher level of expression,as compared with poorly differentiated gastric carcinomas (P<0. 05). There was no significant difference in expression between the carcinomas with and without metastasis. These results suggest that MDR in gastric carcinoma is readily induced by chemotherapy and that higher expression associates with moderate differentiation. MDR should be taken into consideration if chemotherapy for gastric carcinomas is expected effective.
