摘要
用NAG酶释放法检测细胞毒,因常规底物PNP-NAG灵敏度低而无法使用。故使用发荧光底物4Mu-NAG,提高了方法灵敏度,同时对诸如靶细胞数、底物浓度、NP-40的影响,适宜的效靶比等一系列因素进行了检测。确定了最佳反应条件,建立了新方法,并用该方法测出了LAK细胞的活性曲线,与文献对比,结果一致。从而证明NAG酶荧光比色法是测定细胞毒的良好方法。
Because of the low sensitivity of routine sustrate PNP-NAG,NAG enzyme releasing method could not be used in detecting cytotoxicity. In this article,fluorescence substrate 4Mu-NAG was used. Meanwhile,a set of influencing factors such as: the number of target cells,concentration of substrate, influence of NP-40, proper effect-target ratio were studied. After that we have set up a new method with very high sensitivity and the best reactive condition, and using this method the activity curve of LAK cells was depicted. Same result was achieved as compared with the references. It is indicated that NAG enzyme fluorescence analysis is a good method in cytotoxicity detection.
