胃癌发生发展中可溶型Fas/FasL的表达及其意义 被引量:1

摘要 目的:探讨Fas/FasL基因蛋白在胃癌发生发展中的作用。方法:应用免疫组化、特定组织蛋白提取及Westem印迹杂交技术,对20例正常胃黏膜,22例癌前病变和35例胃癌组织中Fas/FasL的表达情况加以分析。结果:正常胃黏膜、癌前病变、胃癌组织的Fas阳性率分别为85%、45%、37%;FasL的阳性率分别为20%、27%、46%。Western印迹杂交显示:胃癌细胞产生的Fas蛋白以分子量30 kDa的可溶型Fas(sFas)为主,伴少量的43 kDa膜型Fas(mFas)。结论:Fas在正常胃黏膜和癌前病变、胃癌组织中的表达强度差异有显著性(P<0.05);FasL在胃癌组织和正常胃黏膜、癌前病变中的表达强度差异有显著性(P<0.05)。 Objective: To investigate the possible role of Fas, FasL in human gastric carcinogenesis. Methods: The expression of Fas and FasL protein was examined by immunohistochemistry staining, protein extraction and Westem blot hybridization. Results: The positive rates of Fas in normal epithelial cells, dysplastic cells and carcinoma cells were 85 %, 45 % and 37 %, respectively. The positive rates of FasL in normal epithelial cells, dysplastic cells and carcinoma cells were 20%, 27% and 46%, respectively. The results of Westem-blotring: the MW of Fas is 30 kDa, and the style of Fas is soluble Fas (sFas). Conclusion: Fas expression is significantly different between normal epithelial cells and dysplastic cells, carcinoma cells ( P 〈 0.05). FasL expression is significantly different between carcinoma cells and normal epithelial cells, dysplastic cells ( P 〈 0. 05).
