摘要
目的:通过体外实验探讨rALR对脾脏单个核细胞是否具有直接的免疫调控作用和作用特点。方法:以3H-TdR掺入法检测脾脏单个核细胞在不同处理情况下的增殖状况:①不同剂量的rALR与5μg/ml ConA同时加入;②5μg/ml ConA预刺激脾脏单个核细胞10和32小时后,再加入30μg/ml rALR;③30μg/ml rALR预处理脾脏单个核细胞至30小时,再加5μg/ml ConA刺激。以环孢霉素A作为阳性对照,以转空质粒的酵母表达上清提取物作为阴性对照。以放免法检测培养上清中IL-2分泌情况。结果:不同剂量的rALR与ConA同时加入时,能以剂量依赖方式明显抑制ConA对脾脏单个核细胞的促增殖作用。ConA预刺激脾脏单个核细胞10、32小时后,再给予30μg/ml rALR仍可明显抑制脾脏单个核细胞的增殖,但用30μg/ml rALR预处理脾脏单个核细胞至30小时,并不影响脾脏单个核细胞对ConA刺激的反应能力。ConA+rALR(30μg/ml)组的IL-2分泌在24小时明显低于ConA组。结论:rALR对ConA活化的脾脏单个核细胞增殖有明显的抑制作用,而对未活化的脾脏单个核细胞则无明显影响,提示未经抗原激活的免疫细胞可能不表达ALR受体。
Objective:To study the possible role and way of ALR in the immune regulation in vitro. Methods:The proliferation of spleen mononuelear cells of rat was detected with 3 H-TdR method in vitro under the following different treatments: ( 1 ) the administration of the different concentration of rALR with 5 μml ConA at same time ; ( 2 ) the addition of 30 μg/ml ALR after 5 μg/ml ConA pretreatment in 10 h and 32 h, respectively; ( 3 ) the addition of 5 μg/ml ConA after 30 μg/ml ALR pretreatment until 30 h. Cyelosporin A and the supernatant from cultures of yeast were used as positive and negative controls, respectively. The IL-2 levels in the supernatants from the mononuelear cells by various treatments were detected with R/A test kit. Results: rALR inhibited the proliferation and the production of IL-2 of the mononuelear cells from rat spleen stimulated by ConA dose-dependently. The mononuelear cell proliferations were still inhibited by 30 μg/ml rALR after stimulated by ConA for 10 h or 32 h, but the pretreatment of 30 μg/ml rALR for 30 h had no influence on the reactivity of mononuelear cell to ConA compared with control. Conclusion:rALR could inhibit in dose-dependent way the proliferation of mononuelear cells from rat spleen stimulated by ConA in vitro, but it eouldn' t influence the rest mononuclear cells. This suggests that the rest mononuelear cells might not express the receptor of ALR.
