摘要
目的:探索泻心汤组方药物不同配伍情况下其中所含的黄酮类、蒽醌类、生物碱类成分的含量变化。方法:采用HPLC-DAD-MS法,以乙腈-0.02mol·L-1醋酸铵溶液(用醋酸调节pH3.50)为流动相进行梯度洗脱,DAD检测波长为254、278和270nm。结果:确定了其中20种化学成分的变化趋势并对10种成分进行了含量测定。结论:大黄、黄芩、黄连三者两两合煎时,其中所含三类成分均存在动态变化;三者合煎时大黄中的蒽醌类成分除大黄酸外均升高,黄芩中的黄酮类成分和黄连中的生物碱类成分其煎出率均有不同程度地降低。HPLC-DAD-MS法可用于研究中药复方配伍后化学成分的变化规律。
AIM: To explore the change of contents of flavonoids, anthraquinones and alkaloids among different compatibilities of medicinal materials in Xiexin decoction. METHODS: By HPLC-DAD-MS, with acetonitrile-0.02mol· L^-1 acetic ammonium( adjusted pH to 3.50 with acetic acid)as mobile phase(using gradient)and the wavelength of the DAD detector was set at 254 rim, 278 nm and 270 nm. RESULTS: The mutative trend of 20 compounds could be clarified by HPLC-MS, among which 10 had been determined by HPLC-DAD. CONCLUSION: The assay results showed that the contents of these compounds were related to different compatibilities. When the three medicinal materials were decocted together, the anthraquinones were increased except Rhein, while the flavonids and the alkaloids were all decreased to some extent. HPLC-DAD-MS could be used to study the compatibility of some traditional Chinese medicines.
