摘要
目的观察寻常型银屑病病人外周血单核细胞Toll样受体2(TLR2)mRNA表达及血清抗链球菌溶血素“O”(ASO)和白细胞介素-8(IL-8)水平的变化,探讨其与银屑病发病的关系。方法采用RT-PCR方法检测银屑病病人(银屑病组)外周血单核细胞TLR2 mRNA表达,采用胶乳凝集法检测血清ASO水平,采用ELISA法检测血清IL-8水平。以我院体检健康者30例作为对照(正常对照组)。结果银屑病组TLR2 mRNA表达明显高于正常对照组(t=5.541,P<0.01);银屑病组点滴型与斑块型TLR2 mRNA表达比较无显著性差异(t=0.318,P>0.05)。银屑病组血清ASO水平明显高于正常对照组(2χ=8.604,P<0.01);且点滴型病人明显高于斑块型病人(χ2=9.780,P<0.01)。银屑病组血清IL-8水平显著高于正常对照组(t=28.463,P<0.01);点滴型与斑块型病人之间比较无显著性差异(t=0.660,P>0.05)。点滴型病人ASO阳性者TLR2 mRNA表达水平高于ASO阴性者,差异有显著性(t=2.407,P<0.05);银屑病病人外周血TLR2 mRNA表达与IL-8水平呈正相关(r=0.637,P<0.01)。结论TLR2可能通过识别链球菌等入侵病原微生物启动机体免疫应答,介导细胞因子IL-8等的产生,参与银屑病皮损的发生和发展。
Objective To investigate the expression of toll-like receptor 2 (TLR2) mRNA in monocytes of psoriatic patients' blood and the serum levels of interleukin-8 (IL-8) and anti-streptolysin O (ASO), and to explore their relevance to pathogenesis of psoriasis. Methods Expression of TLR2 mRNA were detected by reverse transcription polymerase chain reaction (RT-PCR) method ; serum levels of ASO and IL-8 were detected by latex agglutination assay and enzyme linked immunosorbent assay (ELISA) respectively. Results Expression of TLR2 mRNA of the patients was higher than that of the controls (t=5. 541, P〈0.01). There was no significant difference in the expression of TLR2 mRNA between guttate and plaque psoriatic patients (t= 0. 318,P〉0. 05). The positive rate of ASOin the patients was higher than that of the controls (x^2=8.604,P〈0.01); and the ASO positive rate in guttate patients was higher than that in plaque psoriatic patients (x^2 = 9. 780,P〈0.01). Level of IL-8 in psoriatic patients was higher than that controls (t=28. 463,P〈0.01) ; there was no significant difference between guttate and plaque psoriatic patients (t=0. 660,P〉0.05). The expression of TLR2 mRNA in psoriatic patients with positive ASO was higher than that in patients with negetive ASO (t=2. 407 ,P〈0.05) ; the expressions of TLR2 mRNA and IL-8 in psoriatic patients had a positive correlation (r=0. 637, P〈0.01), Conclusion TLR2 may be involved in the occurence and development of psoriasis by recognizing the invading streptosis and the following generation of cytokines.
