摘要
以分光光度法定量,优化香草醛-高氯酸法的显色条件:50 g/L香草醛-冰乙酸与高氯酸分别为0.3 mL和0.6 mL,80℃水浴10 min,测定波长550 nm。以人参皂甙Rb1为对照品,绘制标准曲线,回归方程m=0.162 7A+0.000 1,相关系数r=0.999 6。结果显示,分光光度法测定青钱柳叶中总三萜皂甙含量的精密度、加样回收率、稳定性试验RSD分别为2.28%、2.42%、1.99%。
The content of total triterpene saponins was determined by spectrophotometry, the optimal condition was determined in the vanillin - perchloricacid method : 0.3 mL of 50 g/L vanillin in acetic solution and 0.6 mL of perchloricacid was added, then the mixture was warmed up in a waterbath at 80℃ for 10 min, and the detection wavelenth was 550 nm. And the ginsenoside Rb1 standard sample was usded to make standard curve, the regressive equation was m =0. 162 7A +0. 000 1 ,the correlation coefficient was 0.999 6. The experimental result showed the relative standard deviations (RSD) of the accuracy,recoveries and stability were 2.28% ,2.42% and 1.99% ,respectively.
