氧自由基在家兔低血容量性休克-再灌注损伤中的作用及川芎嗪的保护效应 被引量:23

摘要 目的:探讨氧自由基在家兔低血容量性休克再灌注损伤中的作用,并观察川芎嗪对其有无保护效应。方法:制备家兔低血容量性休克模型,检测血浆和组织超氧化物歧化酶(SOD)活性及丙二醛(MDA)含量。实验分为川芎嗪保护组和未用川芎嗪的非保护组,并进行比较。结果:休克前两组动物SOD活性及MDA值均无统计学差异。休克90分钟时两组动物SOD活性均显著下降(川芎嗪保护组为187±36NU/ml,非保护组为189±37NU/ml),MDA值均显著升高(保护组8.10±0.90μmol/L,非保护组8.10±0.88μmol/L);休克再灌注后,保护组SOD活性逐渐上升,休克再灌注3小时后接近休克前水平(342±29NU/ml),明显高于休克90分钟(187±36NU/ml)和非保护组同时相水平(121±22NU/ml);保护组MDA逐渐下降,休克再灌注3小时后(6.00±0.62μmol/L)明显低于休克90分钟时(8.10±0.90μmol/L)及非保护组同时相水平(9.30±0.96μmol/L)。此外,川芎嗪保护组心、肝、肾、肠道组织SOD活性明显高于非保护组SOD活性;心、肝、肠道MDA含量明显低于非保护组MDA值。? Objective:To investigate the role of oxygen free radicals in reperfusion injury in rabbits following hemorrhagic shock.Methods:Using a hemorrhagic shock model,serum and tissue superoxide dismutase (SOD) and malondialdehyde (MDA) were measured in 20 rabbits.Animals were randomly divided into ligustrazin treatment group (n=10) and nontreatment group (n=10).Results:Baseline measurements of SOD and MDA were similar in two groups.At a 90min hemorrhage,serum SOD activity significantly decreased and MDA concentration increased in both groups as compared to baseline values (all P <0 01 vs.baseline).With treatment of ligustrazin,SOD increased gradually and had a tendency approaching baseline (342±29NU/ml) at 3 hours after reperfusion,which was significantly higher than that of 90min hemorrhage (187±36NU/ml) and nontreatment group (121±22NU/ml).In treatment group MDA decreased after reperfusion,being 6 00±0 62μmol/L at 3 hours,and it was markedly lower than that of 90min hemorrhage(8 10±0 90μmol/L) and nontreatment group (9 30±0 96μmol/L).In addition,SOD activity in the heart,liver,kidney and intesine in ligustrazintreated group were higher than those in nontreated group,while MDA concentration in the heart,liver and intestine in ligustrazintreated group were lower than those in nontreated group.Conclusions:It is indicated that oxygen free radicals may be one of the primary causes for reperfusion injury after hemorrhagic shock.Ligustrazin can attenuate reperfusion injury associated with hemorrhagic shock by eliminating oxygen free radicals.
