模拟失重对大鼠血清及肺组织细胞因子的影响研究 被引量:4

摘要 目的研究模拟失重对大鼠血清中细胞因子(TNF-α,IL-6和IL-8)和肺组织中原癌基因C-fos的影响。方法-30°尾部悬吊(TS)大鼠模拟失重生理效应。40只Wistar大鼠随机分为7d对照组(7dCON)、21dCON、TS7d和TS21d4组。血清细胞因子(TNF-α,IL-6和IL-8)及肺组织中原癌基因C-fos的动态表达分别采用酶联免疫吸附试验法(ELISA)和免疫组织化学法检测。结果TS7d和TS21d大鼠血清中TNF-α,IL-6,和IL-8与对照组(CON)比释放明显增高,差异有统计学意义(P<0.01,P<0.05),且TS7d和TS21d大鼠肺组织C-fos的表达水平与对照组(CON)比差异也具有统计学意义(P<0.01,P<0.05)。结论尾部悬吊模拟失重可能引起大鼠肺脏一定程度的损伤,并对血清炎性细胞因子TNF-α,IL-6和IL-8的释放及肺组织C-fos的表达产生影响。 Objective To investigate the effects of simulated weightlessness on TNF-α IL-6 and IL-8 in serum and C-fos on pulmonary tissues in rats. Methods -30° tail-suspension(TS) rats were used as the model to simulate the physiological effects of weightlessness. Fourty Wistar male rats were randomly arranged into 4 groups: control for 7d (7d CON), 21d CON, TS for 7d (TS7d) and TS21d. The dynamic expresses of C-los in the sera and lung tissues were respectively observed by using ELISA and the immunohisochemical technique. Results The contents of TNF-α, IL-6 and IL-8 in sera of 7d and 21d tail-suspensed rats increased significantly(P〈0.01, P〈0.05), while the activation level of C-fos in lung tissue of 7d and 21d tail-suspensed rats increased significantly(P〈0.01,P〈0.05). Conclusion TS resulted in some lung injury of rats, which could have some effects on the pulmonary circulation and function.
