定量检测多药耐药基因在胃癌、大肠癌组织中的表达及其临床意义 被引量:7

摘要 应用逆转录多聚酶链式反应(RT-PCR)技术,定量检测了68例胃癌,58例大肠癌组织中多药耐药基因(MDR1)的表达水平,并对11例胃癌及大肠癌患者进行了追踪检测。结果表明:化疗后肿瘤组织细胞中MDR1的表达量(>0.5)明显高于化疗前(<0.5),MDR1的表达量化疗后与化疗前比较差异显著(P<0.01),而且随化疗疗程数的增加,MDR1表达量有上升的趋势。患者化疗效果与MDR1的表达密切相关,与MDR1的表达量成反比。MDR1的表达可成为临床医生监测病情、选择有效的化疗药物、判断预后的重要参考指标。此法以临床标本为样品直接进行检测,以MDR1/β2M为MDR1的表达量,排除了实验过程中诸多因素的干扰,使样本之间的比较更加合理,结果准确、可信。 We have devised a highly sensitive, specific and quantitative protocol for measuring the levels of MDR1 mRNA in human gastrointestinal tract carcinomas, based on RT PCR. The relationship between the expression of MDR1 gene and clinical chemotherapy of gastrointestinal carcinomas has been analyzed. The results show that low levels of MDR1 expression were found in untreated cancers and high levels in the treated cancers. The levels of MDR1 expression had a significant association with the clinical chemotherapy(P<0.01). And the epression of MDR1 gene had been elevating with the increasing of chemotherapy. We have followed 11 patients to detect the level of MDR1 mRNA. We found that the expression of MDR1 gene is closely related to results of treatment. Our assay shows that MDR1 mRNA expression may be a useful indicator for evaluating the prognosis of gastrointestinal tract carcinomas.
