血浆中吡格列酮的测定及其在人体内的药动学研究 被引量:2

摘要 目的建立测定血浆中吡格列酮的方法。方法采用Diamosil C18(150 mm×4.6 mm,5μm)色谱柱,以乙腈-pH5.8磷酸盐缓冲液(55:45)为流动相,安定为内标,检测波长225 nm。结果吡格列酮在0.02-2.00μg·ml^-1范围内,线性良好(r=0.9968),平均回收率为105.3%,日内、日间的RSD为4.23%-18.40%。结论所建方法操作简便、快速、重复性好、准确可靠,适用于临床血药浓度的监测及药动学研究。 OBJECTIVE To establish a method for determination of Pioglitazone in human plasma. METHODS Using HPLC for determination of Pioglitazone in human plasma, the Diamosil C18 column was taken as analytical column, the mobile phase consisted of acetonitrile -phosphate buffer (55:45 ) , flow rate was 1.0 ml·min^-1. The internal standard was Diazepam (2 μg·ml^-1 in methanol). The detection wavelength was 225 nm. RESULTS The linear range of Pioglitazone was 0.02 - 2. 00 μg·ml^-1( r = 0.9968 ). The average recovery was 105.3% with the RSD of 7.09%. The intra day and inter - day precisions expressed as RSD ranged from 4. 23% - 18.40%. CONCLUSION The HPLC method is quick, accurate, precise and repeatable. It can be used to determine the concentration of Pioglitazone in human plasma and and spplied in pharmacokinetic studies.
