不同浓度硫化氢对离体大鼠心脏低温保存效果的研究 被引量:3

Protective effects of hypothermic St.ThomasⅡcardioplegia solutions with different concentrations of H_2 S on isolated rat hearts

摘要 目的探讨大鼠离体心脏在含有各种浓度硫化氢(H_2S)的HST(H_2S St.ThomasⅡ)液中的的低温保存效果。方法将32只SD大鼠随机平均分为4组,分别用Ⅰ组(对照组):托马斯(St.ThomasⅡ,STH)液;Ⅱ组:含4×10^(-2)mmol/L硫氢化钠(NaHS)的HST液;Ⅲ组:含4×10^(-1) mmol/L NaHS的HST液;Ⅳ组:含4 mmol/L NaHS的HST液对心脏进行保存。采用Langendorff心脏灌注和工作模型装置进行心脏功能测定,比较6 h后心功能恢复率以及能量变化、心肌含水量,观察心肌超微结构改变。结果心脏保存6 h后,Ⅳ组左心室功能损伤严重,Ⅱ、Ⅲ组左心室功能损伤较轻,其中Ⅲ组最轻。Ⅲ组冠状动脉流量(CF)恢复率为(88.05±8.44)%,明显比Ⅰ组(73.17±8.55)%、Ⅳ组(76.95±9.44)%好,差异有统计学意义(P<0.01);Ⅰ组心肌中ATP、乳酸(LD)、糖原含量及心肌含水量分别为(4.31±1.34)μmol/g、(28.70±2.32)μmol/L、(23.00±0.32)mg/g、(79.74±5.47)%,与其他3组比较,差异有统计学意义;Ⅳ组心脏复跳时间为(113.38±17.71)s,明显较Ⅰ组(63.38±9.20)s为长(P<0.01);心肌超微结构Ⅲ组损伤最轻;Ⅲ组心肌保存良好。结论HST保存液对大鼠心脏的保存在6 h内有明显的保护作用,效果优于STH液;含4×10^(-1)mmol/L NaHS的HST液保存效果较好,而高浓度硫化氢对心肌有损害作用。 Objective To evaluate the protective effects of a novel hyperpolarized cardiac preservation solution with different concentrations of H2 S on isolated rat hearts. Methods Thirty-two SD rats were randomly divided into 4 groups : In group Ⅰ donor hearts was preserved in pure STH ( St. Thomas Ⅱ ) cardioplegia solution,in group Ⅱ in HST solution containing 4 × 10^-2 mmol/L NariS,in group Ⅲ in HST solution containing 4 × 10^-1 mmol/L NariS, and in group Ⅳ in HST solution containing 4 mmol/L NariS and their hearts were preserved respectively for 6 h. The isolated hearts were perfused in the Langendorff rat mode for functional evaluation before and after preservation. ATP, LD, glycogen, wet weights in myocardium and cardiac uhrastructure were observed after reperfusion. Results The impairment of the left ventricle function in group Ⅳ was serious, while that in group Ⅲ was mild. The recovery of coronary flow in group Ⅲ was (88.05 ±8.44)% ,significantly higher than the other three groups (P 〈0.01 ). The contents of ATP, LD, glycogen and wet weight of myocardium in group Ⅰ were (4.31 ± 0.34) μmol/g, (28.70 ± 2.32 ) μmol/L, (23.00 ± 0.32 ) mg/g and (79.74 ± 5.47 ) % respectively. There were significant differences in the contents of ATP, LD, glycogen and wet weight of myocardium between group Ⅰ and the other three groups. The time from the reperfusion to beating of heart was longest in group Ⅳ. The ultrastructure changes were milder in group Ⅲ. Conclusion HST solution can protect rat heart for 6 h effectively. The concentration of 4 × 10^-1 mmol/L NariS has better preservation effect and HST eardioplegia solution is more effective than STH eardioplegia solution. High concentration of H2S solution for myocardial preservation is harmful to heart.
