摘要
目的:观察来氟米特对单侧输尿管梗阻大鼠肾间质纤维化指标PCNA、α-SMA及ColⅠ、ColⅢ的表达程度的影响及相互关系,从而探讨其作用机制。方法:将36只健康、雄性SD大鼠中的24只行左输尿管结扎术,另外12只行假手术。术后第2天开始给药,结扎后的大鼠分为UUO组和治疗组。术后第7、14天分别处死各组中的6只大鼠,用免疫组化方法测定PCNA、α-SMA、ColⅠ、ColⅢ的表达情况。行HE和Masson染色,动态观察肾间质病理学改变。结果:来氟米特能显著减少UUO大鼠肾小管间质的PCNA、α-SMA表达,减轻ColⅠ和ColⅢ在肾小管间质的沉积。结论:来氟米特可抑制受伤的肾小管间质α-SMA的表达,减少基质的聚集从而减轻肾间质纤维化。
Objective: To study the effect of leflunomide (Lef)on the process of renal interstitial fibrosis caused by unilateral ureteral obstruetion(UUO) in rats. Methods: 24 Sprague-Dawley rats underwent UUO and were randomly divided into two groups: UUO group (n=12) and Lef-treated group ( = 12). Then they respectively received vehicle or Lef(10 mg/kg· d-1 by daily gastric gavage)during the 14 days following surgery. Additional 12 rats received sham operation. Six rats of each group were killed respectively in 7 and 14days after surgery. Immunohistoehemistry was performed on renal tissue for proliferating cell nuclear antigen(PCNA),α-smooth muscle aetin(α-SMA),which was eytosheletal marker of Myoibroblast(Myo) and collagen I , Ⅲ. Histological changes were also observed by HE and Masson stain. Result: In comparison with the UUO group, α-smooth muscle aetin, PCNA and collagen I , Ⅲ deposition were all significantly reduced by Lef treatment. Conclusion: Lef can prevent renal interstitial fibrosis process in UUO rats by reducing interstitial Myo infiltration and collagen I ,Ⅲ deposition. Lef may be used to slow the progression of chronic renal disease.
