摘要
从海洋中筛选出1株产SOD能力较强的酵母菌菌株Y9,研究了不同碳源、氮源、碳氮比和金属离子对其细胞生物量及产SOD的影响。该菌株产SOD的适宜碳源为葡萄糖,氮源为蛋白胨,氮源浓度为2%,适宜金属离子为Mn2+。以其为出发菌株,通过正交试验优化其产SOD的发酵条件,结果表明,菌株Y9产SOD最适发酵条件为pH6.5、Mn2+浓度150μmol/L、装液量为60mL/500mL、接种量为10%,在此优化条件下,菌株Y9的SOD总量可达2280.2U/L,比初始产量提高了14.2%。
The SOD high-producing strain Y9 of yeast was screened from marine. The effects of carbon sources, nitrogen sources, ratio of carbon to nitrogen and metallic ion on biomass and SOD activity of the strain Y9 were studied in the paper. The results showed that the optimal carbon source was glucose, the optimal nitrogen source was peptone, the optimal nitrogen concentration was 2% and the optimal metallic ion was Mn^2+ for Y9 SOD producing. The optimal conditions for SOD fermentation were studied with orthogonal test. The results showed that the optimal conditions were as follows: pH6.0, Mn^2+ 150μmol/L, volumes 60mL/500mL and inoculation 10%. The SOD yields of strain Y9 could be 2280.2U/L in optimal medium and 14.2% were higher than the first separation yields.
