The Effect of Noradrenalin on the Expression of Vascular Endothelial Growth Factor in Astrocytes

摘要 目的:探讨去甲肾上腺素(Noradrenalin,NA)对星形胶质细胞血管内皮生长因子(Vascu-larendothelial growth factor,VEGF)表达的影响,阐明NA促进高原脑水肿形成的机制。方法:建立大鼠胶质细胞培养模型。分为正常对照组、LNA组、MNA组及HNA组,分别在培养液中加入0μM、5μM、10μM、30μM NA(Sigma公司,美国),在常氧培养条件下(5%CO2、95%O2)培养2h(提取总RNA)或20h(提取总蛋白质)。应用SYBR(Green Quantitative RT-PCR试剂盒(Sigma公司,美国)在RG-3000仪上进行VEGF-mRNA荧光定量RT-PCR检测;应用蛋白印迹法检测VEGF的表达。结果:与对照组相比,LNA组、MNA组及HNA组星形胶质细胞VEGF-mRNA的表达持续显著增高(分别P<0.01,P<0.01,P<0.001),与NA浓度呈剂量依赖关系。与对照组相比,LNA组星形胶质细胞VEGF表达有一定程度升高,但无差别显著性(P>0.05),而MNA组及HNA组星形胶质细胞VEGF的表达量显著增高(分别P<0.01,P<0.001),与NA浓度呈剂量依赖关系。结论:NA可从分子水平诱导星形胶质细胞表达VEGF,呈剂量依赖关系。 Objective: To investigated the effect of noradrenalin on the expression of vascular endothelial growth factor (VEGF) in astrocytes to elucidating the mechanism of NA in the formation of high altitude cerebral edema. Methods: The hypoxic primary astrocyte culture model was established. Various levels of noradrenalin (0μM as the normal control group,5μM as LNA, 10μM as MNA,30μM as HNA) were given to induce VEGF--mRNA and VEGF expression. The astrOcytes were processed for real--time quantitative RT--PCR to detect expression of VEGF--mRNA and Western blots to detect VEGF expression. Results: In normal control group, the VEGF--mRNA expression was very low. There was significant difference in VEGF--mRNA expression between normal astrocytes and those treated with LNE (P〈0.01). After treating with MNE and HNE, the expression of VEGF--mRNA increased more obviously (P〈0. 01, P〈 0. 001 respectively). In normal control group, the VEGF expression was very low. There was no significant difference in VEGF expression between normal astrocytes and those treated with LNE. After treating with MNE and HNE, the expression of VEGF increased obviously(P〈0. 01, P〈0. 001 respectively). Conclusions:The results suggested that noradrenalin may induce astrocytes to express VEGF--mRNA and VEGF in a dose--dependent manner, which could participate in the formation of high altitude cerebral edema.
