摘要
选用特早熟杏红荷包为试材,首次将果实发育期45天的胚培养成株,从发育55~59天的胚得到300余株壮苗,栽植于选种圃.提高特早熟杏胚培成首的技术关键主要是:①选择适宜的接种时期,在花后59天硬核期PF1值达0.9时采样接种;②选用改良Tuber培养基,附加BAO.2毫克/升;③经80天以上0~3℃低温处理,④在15℃条件下,增强光照炼苗,9月下旬气温较低时移栽.
More than 300 vigorous plantlets have been got by culture of 55-59 d embryos from the ear-liest apricot variety Honghebao and transplanted in a breeding nursery. For the first time someplantlets have been cultured from the embryos at 45 d. The key techniques to increase percentageof plantlet formation were (i) sampling at 59 d after blossoming when the embryo was at stone-hardening stage with PF, being 0. 9, (ii) using the improved Turkey medium supplemented withBA (0. 2mg L-1), (iii) culturing at O-3℃ for more than 80 d, (iv) training at 15℃ with gradual-ly increased illumination, then transplanting to field in late September.
