肺癌细胞系线粒体DNA非编码区序列变异研究 被引量:2

摘要 背景与目的线粒体DNA(mtDNA)非编码区在mtDNA转录过程中起重要的调控作用,mtDNA非编码区的突变将直接影响其转录和翻译,从而改变细胞的能量代谢。本研究的目的是分析7种肺癌细胞系mtDNA非编码区序列的变异情况并探讨其意义,为肺癌细胞系mtDNA变异研究提供基础信息。方法培养7种常用肺癌细胞系,包括人肺腺癌A549、SPC-A-1、Calu-3、LTEP-a-2,人肺扁平上皮癌QG-56,人高转移肺癌95-D以及人小细胞肺癌NCI-H446,应用改良一步法提取7种肺癌细胞系mtDNA,以剑桥mtDNA序列为标准,分析非编码区序列变异情况。结果7种肺癌细胞系mtDNA非编码区存在不同程度的碱基变异,其中LTEP-a-2细胞系变异率最高,为1.82%,A549细胞系最低,为0.21%。结论率先成功完成了7种人肺癌细胞系mtDNA整个非编码区的测序,揭示了这7种肺癌细胞系mtDNA非编码区碱基变异背景,为后续实验研究提供了理论基础和参考依据。 Background and objective It has been proven that mitochondrial DNA (mtDNA) noncoding region plays a very important role in process of transcription. The mutation in mtDNA noncoding region can influence the transcription and translation process of mtDNA. The aim of this study is to investigate the variation status in noncoding region of mtDNA in lung cancer cell lines, and discuss its meaning in carcinogenesis. Methods The 7 lung cancer cell lines, A549, SPC-A-1, Calu-3, LTEP-a-2, QG-56, 95-D and NCI-H446, were cultured. Then the mtDNA of them was extracted with one-step method and variation status of noncoding region was analyzed, compared to the Cambridge sequence of mtDNA. Results Different variations existed in the mtDNA noncoding regions of 7 lung cancer cell lines. The variation rate of LTEP-a-2 (1.82%) was the highest one, and A549 (0.21%) was the lowest one. Conclusion The mtDNA noncoding region sequencing is accomplished firstly in 7 lung cancer cell lines. It may provide a reference and background for related researches.
