脐带血干/祖细胞的体外扩增实验研究进展 被引量:2

摘要 在造血干细胞移植中,脐带血造血干细胞以其独有的特性,成为骨髓及外周血造血干细胞的极佳替代来源。但是单份脐带血所含有的造血干/祖细胞(HSPC)数目相对少,不能满足大体重儿童及成年人需要。采用联合细胞因子、基质细胞共培养或者灌注培养等方法,可以在体外对脐带血HSPC进行扩增,有望解决这一难题。其可能的机制在于外源性的生长因子能阻止细胞凋亡,刺激细胞增殖;基质细胞能维持HSPC的自我更新能力,同时能促进植入以及调节免疫;灌注培养方式能维持稳定的培养环境,及时补充营养物和移出代谢物,从而有利于细胞的扩增。在扩增细胞的同时应尽量减少干细胞的分化,维持其长期造血能力。该文对HSPC的生物学特点,扩增研究的进展及其临床应用进行综述。
