摘要
目的:观察大鼠弥漫性脑损伤后BDNF的表达变化。方法:采用Marmarou的弥漫性脑损伤模型,分别于伤后15min、30min、lh、3h、6h、12h、24h、48h、72h取脑组织,运用免疫组化SABC法观察BDNF的表达,以正常大鼠和假手术大鼠作为对照。结果:正常大鼠即有少量BDNF的表达。打击后15min,BDNF表达增强,1h达高峰,48h降至正常水平。结论:机体对脑损伤存在自我保护机制;BDNF的时序性变化可望为法医学脑损伤时间的推断提供参考。
To investigate the expression of BDNF in diffuse brain injury ( DBI ) in rats. Methods : Based on Marmarou' s model of DBI, the brain tissue samples were taken at 15rain,30min,1h,3h,6h, 12 h,24h,48h and 72h respectively after brain injury. The expression of BDNF was examined by immunohistochemical SABC approach and compared with normal control and sham-operation control group. Results: In the brain of normal control and sham operation control groups,the low expression level of BDNF was observed. The expression of BDNF increased at 15 min after injury,peaked at 1h and returned to basal level at 48h. Conclusion : The findings suggest that the organism produces self-protection against brain injury. The level change of BDNF with injury time might provide evidence for detecting brain injury.
