摘要
分别以芹菜种子、子叶、胚轴、叶柄和叶片为外植体,在加有2.4-D和KT的MS培养基诱导下,产生的大量胚性愈伤组织,继代两年后仍有高频再生植株的能力。从胚性愈伤组织建立的悬浮培养细胞系中分离到的单细胞,经4种方法培养后,均得到愈伤组织,并能高频再生植株。在细胞培养过程中,可观察到少数单细胞进行不均等分裂,形成类似于原球胚的结构,从而进一步证明了体细胞胚可以起源于单细胞。
Seeds, cotyledons, hypocotyls, petioles and leaves of Chinese Celery (Apiumgraveolens L.) were used as explants to induce callus on a solid MS medium supple-mented with 1ppm 2.4-D and 0.5 ppm KT. Somatic embryogenesis remains in highpercentage after subculturing two years. Embryogenic callus was cultured in suspen-sion in B_5 medium supplemented with 1 ppm 2.4-D to isolate single cells. Singlecells were cultured with four different methods and all produced calli. Whole plantswere regenerated from these calli in high percentage. It was concluded that a singlecell can form the structure like proembryo by continuous division, and that somaticembryo can be formed from a single cell.
