中华蜜蜂化学感受蛋白cDNA克隆、定位及其表达 被引量:6

摘要 化学感受蛋白被认为是化学通讯中广泛存在于感觉器官中的一种小分子蛋白.在中国本土蜜蜂中华蜜蜂Apis cerana cerana工蜂的触角中,克隆并鉴定了一种化学感受蛋白,命名为Ac-ASP3.Ac-ASP3全长cDNA由2个外显子组成,可读框长393bp,编码130个氨基酸.序列分析表明,其具有保守的4个半胱氨酸,并在N端具有19个氨基酸的信号肽.另外,Ac-ASP3与其同一家族基因意大利蜜蜂Apis mellifera的Am-ASP3具有较高的相似性,而与其他物种相似性较低.免疫定位结果显示,Ac-ASP3仅特异性地表达于机械感器,如毛形感器B和锥形感器中;而在嗅觉感器,如板形感器中几乎无分布.实时定量PCR结果表明,Ac-ASP3高丰度表达于翅和足中,而在触角中表达量却相对较低.在时间表达上,Ac-ASP3在蛹期及成虫第1~6日龄阶段大量表达,这一时期中华蜜蜂正处于幼年期,并在蜂巢进行日常清洁和哺育幼虫等活动.以上结果表明,Ac-ASP3是具有种群特异性的,不参与搜寻蜜粉源等嗅觉活动,但可能参与蜂巢内个体信息识别以及触角的机械运动.
