下颌骨牵引成骨过程中转化生长因子表达 被引量:3

Expression of TGF-β1 in mandibular distraction osteogenesis

摘要 目的通过建立兔下颌骨牵引成骨实验动物模型,研究下颌骨牵引成骨过程中TGF-β1表达及意义。方法选用新西兰白兔30只,随机选取6只动物作空白对照组,24只动物右侧下颌骨植入外置式颌骨牵引器。经7 d延迟期后,按1 mm/d速率延长下颌骨7 d,固定期8周。在延迟期第7天,牵引期第2、4、7天,固定期第1、3、6、8周分别处死3只动物,采用免疫组化、RT-PCR方法,检测TGF-β1表达变化。结果TGF-β1表达贯穿下颌骨牵引成骨全过程,下颌骨牵引成骨过程中不同时相多种组织细胞参与表达TGF-β1。TGF-β1mRNA及其蛋白表达在牵引早期达到高峰,此后逐渐减弱。结论TGF-β1在下颌骨牵引成骨血运重建及新骨生成过程中发挥重要作用。TGF-β1可能与传导机械牵引力刺激的细胞信号传导有关,从而启动血运重建和新骨生成机制。 Objective To study the expression of transformating growth factor-β1 (TGF-β1) during mandible distraction osteogenesis(MDO) in a novel experimental model of rabbits. Methods Six rabbits were chosen to be control group, and twenty-four rabbits chosen for experiment were underwent unilateral mandibular distraction (protocol: 7 days latency period, a distraction at speed of 0.5 mm per 12 hours for 7days,and a 8-week consolidation period). Samples were collected at 7 days of latency, 2nd, 4th, 7th day of distraction, and at 1st, 3rd, 6th, 8th week of consolidation. Expression of TGF-β1 and TGF-β1 mRNA were detected by immuno histochemistry and reverse transcription polymerase chain reaction(RT-PCR). Results TGF-β1 were detected in many kinds of cells in distracted area during each time points of MDO. TGF-β1 and TGF-β1 mRNA began to express at early distraction period, peaking at I32, then decreasing slowly. Conclusion TGF-β1 may play an important role in angiogenesis and osteogenesis during MDO. Release of TGF-β1 may act as biochemical signal messengers for the mediation of mechanical stimulus.
