摘要
从华重楼(Paris polyphylla chinensis Franch)的地下块茎中分离出107株内生菌,经TLC筛选检测,菌株RZ03和RZ07可产生薯蓣皂甙。进行了形态和生理生化鉴定并将菌株的16S rDNA序列分别与用BLAST调出GenBank+EMBL+DDBJ中的相关序列比较,用ClustalW绘制系统发育树,RZ03与DQ019167(Exiguobac-terium acetylicum)同源性为99%,鉴定为Exiguobacterium acetylicum,RZ03、RZ07与DQ207730(Bacillus subtilis)序列同源性为99%,初步鉴定为Bacillus subtilisRZ07。
107 endophytes were isolated from underground stem tubers of Paris polyphylla chinensis. RZ03 and RZ07 could produce diosgenin were screened through TLC testing. Their morphology, physiology, and biochemistry were characterized and their 16S rDNA sequence were compared with related sequences obtained from GenBank + EMBL + DDBJ, and phylogenetic tree of their 16S rDNA sequence were drawn with ClustalW respectively. The homology between RZ03 and DQ019167 (Exiguobacterium acetylicum) was 99% , therefore, RZ03 was identified as E. acetylicum RZ03, And The homology between RZ07 and DQ207730 (Bacillus subtilis) was 99% , therefore, RZ07 was identified as B, subtilis RZ07.
