摘要
目的探讨牛磺酸对模拟高原缺氧大鼠小肠组织HIF-1α及HIF-1β表达的影响。方法动物经基础饲料或添加牛磺酸饲料喂养7d后,利用低压舱分别模拟4500m及5500m海拔高度高原缺氧,分别在高原缺氧处理后第2、6、12、24、48、72h处死动物,剪取小肠组织,RT-PCR检测HIF-1α、HIF-1β mRNA表达,Western blot检测HIF-1α蛋白表达。结果缺氧条件下HIF-1α、HIF-1β表达明显增强,在缺氧12h时达到峰值,随后逐渐减弱;添加牛磺酸组在各时相点的表达相对于未加牛磺酸组显著增强。结论牛磺酸能够显著增强高原缺氧大鼠小肠组织HIF-1α、HIF-1β的表达。
Objective To observe the effect of taurine on expression of HIF-1 α and HIF-1 β in the intestinal tissue of rats exposed to hypobaric hypoxia. Methods Totally 208 adult SD rats were randomly divided to raise at 500-meter, simulated 4 500-meter and simulated 5 000-meter altitude, and subdivided by forage: taurine supplementation (intervention) or normal forage (control). After taurine nutritional intervention for 7 d, the rats were respectively exposed to 500-meter, simulated 4 500-meter and 5 500-meter altitude. The expressions of HIF-1α and HIF-1β in the intestinal tissues were determined by RT-PCR and Western blotting in 2, 6, 12, 24, 48, 72 h after exposure to hypoxia. Results After exposure to hypoxia simulated in hypobaric chamber, the expressions of HIF-1α and HIF-1β in intestinal tissues increased obviously, peaked at 12 h after hypoxia, then declined. They were higher in taurine supplementation rats at each time point than that in control rats. Conclusion Taurine nutritional intervention can increase the expressions of HIF-1 α and HIF-1 β in the intestinal tissues of rats exposed to simulated high-altitude hypoxia.
