佐米曲普坦的羟丙基-β-环糊精毛细管电泳手性分析 被引量:5

Enantiomeric Separation of Zolmitriptan by CE with Hydroxypropyl-β-cyclodextrin as Chiral Selector

摘要 建立了毛细管电泳法检测手性药物佐米曲普坦中其(R)-对映异构体杂质。以含30mmol/L羟丙基-β-环糊精的25mmol/L磷酸二氢钾溶液(磷酸调至pH2.0)为运行缓冲液,石英毛细管柱,检测波长为220nm。佐米曲普坦与其对映异构体的分离度为3.5,后者浓度在1~10μg/ml范围内线性良好(r=0.9998),平均加样回收率为100.3%,检测限为0.31μg/ml。 A practical chiral capillary electrophoresis method was established for the determination of the (R)-enantiomer in zolmitriptan. The background electrolyte was 25mmol/L potassium dihydrogen phosphate solution (adjusted to pH 2.0 with phosphoric acid) containing 30mmol/L HP-13-CD. A fused-silica capillary was used at the detection wavelength of 220nm. Zolmitriptan and its (R) -enantiomer were resolved in baseline with resolution of 3.5. The calibration curve of (R) -enantiomer was linear in the concentration range of 1 - 10μg/ml (r = 0.9998). The average recovery was 100.3 % and the detection limit was 0.3 μtg/ml.
