摘要
目的研究碘过量摄入对小鼠仔鼠脑α-tubulin基因表达的影响及硒的干预作用。方法将60只BALB/c小鼠随机分为四组正常对照组(饮用自来水,NC)、碘过量组(饮水含碘3000μg/L,EI+)、补硒组(饮水含Se200μg/L,Se+)、碘过量加硒(饮水含碘3000μg/L+Se200μg/L,EI+Se+)组。以纯系鼠饲料饲养。4个月后,雌雄交配。测定D14和D28仔鼠血清总T4和总T3水平,分别用实时荧光定量PCR和Western Blot测定D0、D14和D28仔鼠大脑组织α-微管蛋白和α-微管蛋白mRNA表达。结果D14仔鼠血清T4水平碘过量组显著低于对照组和碘过量加硒组(P<0.05)。D0仔鼠脑组织α-微管蛋白mRNA水平碘过量组显著低于对照组、单独补硒组和碘过量补硒组(P<0.05)。14d仔鼠脑组织α-微管蛋白mRNA和蛋白表达水平碘过量组显著高于其它组(P<0.05)。碘过量和补硒对D28仔鼠血清T4水平和脑组织α-微管蛋白表达水平无明显影响。结论碘过量引起仔脑α-微管蛋白的发育迟滞,补硒具有缓解作用。
Objective: To study the effects of excess iodine intake and the intervention of selenium on α-tubulin expression in the cerebrum of filial mice. Methods: Sixty BALB/c mice were divided randomly into four groups; control group (tap water, NC), iodine excess group (3 000μg/L I, EI+), selenium group (200μg/L Se, Se+) and iodine excess plus selenium (3 000μg/L I + 200μg/L Se, EI+Se+) group. The mice were mated at the end of the 4 th month. Serum T4 and T3 were determined on postnatal D 14 and D 28. The expression of α-tubulin in filial cerebrum were measured by fluorescent real time PCR and Western blot respectively. Results: Serum T4 in EI+ was lower significantly than that in NC and EI+Se+ on postnatal D 14. mRNA and protein of α-tubulin in EI+ were lower than those in other groups on postnatal D0, but higher on postnatal D 14 (P〈0.05). Serum T4 and α-tubulin among 4 groups were insignificantly different on postnatal D 28.Conclusion: The expression of α-tubulin is lowered in filial cerebrum after iodine excess intake and can be alleviated by selenium supplementation.
