摘要
采用基因工程技术,构建出前列腺组织特异性的基因载体新型C3(1)。利用该载体和转基因技术,培殖出人bcl2基因的前列腺组织特异性转基因鼠。用RNA酶保护法和Western杂交检测发现,雄鼠中转基因人bcl2的mRNA和蛋白只在前列腺和睾丸中有表达。RNA组织原位杂交检测证实人bcl2在鼠前列腺上皮细胞中的过量表达。该实验构建和检测了新型C3(1)作为前列腺组织特异性的基因启动载体的可行性,为前列腺疾病基因治疗研究打下了基础。
By a series of recombinant DNA procedures, we have created an modified C3(1) promoter that was able to directing prostate specific expression of the human bcl 2 cDNA in C3(1) bcl 2 transgenic mice. The transgene expression was confirmed strictly in prostate and testis by RNase protection assay and Western blot analysis.Moreover, in situ hybridization assay found that the expression of transgene was located in prostatic epithelium. The result suggested that the modified C3(1)promoter could target heterologous gene expression specifically to prostate in a developmentally and hormonally regualted fashion, especially could benifit the gene therapy of prostate neoplasia.
