摘要
目的:采用荧光定量-聚合酶链反应(FQ-PCR)法,检测麻疹病毒核酸,用以建立和推广一种敏感、特异、快速的麻疹检测新技术。方法:TaqMan特异性荧光标记探针法,进行特异性、敏感性和麻疹疑似患者临床标本的检测。结果:FQ-PCR法检测麻疹病毒核酸具有高度的特异性和敏感性,本次试验检测灵敏度达0.1 TCID50。在14份麻疹患者咽拭子标本中,有13份标本检出麻疹病毒核酸,从核酸提取至完成检测仅需3 h。结论:FQ-PCR法为麻疹病毒核酸检测提供了一种可靠的方法。
Objective:To establish a specific and sensitive method of FQ - PCR assay for rapid detection of measles virus RNA. Methods:The measles virus RNA was detected by TaqMan FQ - PCR. Results: The sensitivity of this assay was 0. 1 TCID50. Among 14 throat swabs samples, 13 were positive for measles virus RNA. It took only 3 hours to complete the whole course of reaction including extraction of viral RNA and the real -time PCR. Conclusion:This FQ -PCR provides a more efficient method for measles virus RNA detection.
