白细胞介素12对^(60)钴照射小鼠造血功能的影响 被引量:6

The effect of interleukin(IL)12 on hemopoiesis in irradiated mice

摘要 研究白细胞介素12(IL-12)对机体造血系统的调控作用。方法将IL-12两亚基的cDNA分别克隆入不同的真核表达载体,并导入同一NIH3T3成纤维细胞,获得分泌IL-12的基因修饰细胞,经胶原包裹后注射入受照射的小鼠腹腔,观察其分泌的IL-12对小鼠造血功能指标的影响。结果IL-12可以明显增加受照射小鼠的内源性脾结节的数目。7.0Gy照射后10天,空白对照组、母本细胞(3T3-0)组、及分泌IL-12的两个细胞(3T3-C1、C2)组所成脾结节数分别为0.3±0.5、0.5±0.5、8.2±3.3和3.5±0.7。组织学观察显示,7.0Gy照射小鼠中,空白对照及3T3-0组小鼠骨髓腔内造血细胞几乎全部被脂肪细胞代替,仅残余少量明显变性的成熟细胞,而3T3-C1、C2组出现大量的造血细胞,约占骨髓腔总面积的1/15~1/3,并且涉及红系、粒系、巨核系各分化阶段,细胞分裂相多见。脾脏切片显示空白对照及3T3-0组白髓内脾小体数目减少,体积缩小,淋巴细胞严重减少甚至消失,红髓内造血细胞完全消失。而3T3-C1、C2组脾小体数目及大小均明显高于对照组,红髓中出现包括各谱系的造血灶。结论IL-12对受照射小鼠的造? Objective To investigate the regulating effect of IL 12 on hemopoiesis.Methods cDNAs coding for the two subunits of murine IL 12 were cloned into two different eukaryotic expression vectors and transduced into asame NIH3T3 cell line,resulting in two engineered fibroblast clones secreting IL 12. Cells were mixed with collagens and inoculated into the peritoneal cavities of whole body irradiat edmice.Result On day10after treatment,endogenous splenic colony forming unit(CFU S)counts in mice treated with IL 12 secreting NIH3T3 cells were higher than those of mice accepting parental cells or untreated irradiated mice.This difference was statistically significant in both6.0Gy irrdiated mice and 7.0 Gy group mice.Histopathological studies showed that NIH3T3 secreted IL 12 alleviates the injuries caused by irradiation in both bone marrow and spleen,activates hemopoietic cells,increases the number and scale of hemopoietic clusters,hence enhancing hemopoietic function including erythro erythropoiesis,granulopoiesis,and megakaryocytopoiesis.Conclusion IL 12 up regulates and promotes the recovery of hemopoietic function in irradiated mice.
