摘要
目的:研究白藜芦醇经蛋白激酶G对正常豚鼠心室肌细胞L-型钙电流(ICa-L)的影响。方法:采用全细胞膜片钳技术,记录给予白藜芦醇前后ICa-L的变化,并分别记录给予蛋白激酶G(PKG)特异性激动剂8B r-cGMP(100μmol.L-1)、PKG特异性拮抗剂H8(5μmol.L-1)后白藜芦醇对ICa-L的影响。结果:(1)白藜芦醇呈浓度依赖性抑制ICa-L,1、50、100μmol.L-1可使ICa-L峰值分别低18.31%±0.68%,56.20%±2.19%,84.51%±2.52%(n=5,P<0.05),但对ICa-L激活电位、峰电位、反转电位均没有影响;(2)100μmol.L-18B r-cGMP轻度抑制ICa-L,电流密度低10.50%±1.11%,100μmol.L-18B r-cGMP+50μmol.L-1白藜芦醇使ICa-L电流密度降低87.58%±3.49%(n=6,P<0.05);(3)5μmol.L-1H8对ICa-L无影响,5μmol.L-1H8+50μmol.L-1白藜芦醇对ICa-L无抑制作用。结论:白藜芦醇浓度依赖性抑制豚鼠心室肌细胞ICa-L,此作用机制可能与PKG激活有关。
AIM: To investigate the effects of resveratrol on the L - type calcium current in isolated guinea pig ventricular myocytes. METHODS: The whole cell patch clamp method was used. RESULTS: (1) Resveratrol (1, 50, 100 μmol/L) reduced the ICa-L by 18.31%±3.15%, 56.20%±2.50% and 84.51%±4.01% in a concentration - dependent manner ( n = 5, P 〈 0. 05 ). But it has no change on I - V shape of ICa-L. (2) 8Br - cGMP (100 μmol/L), an activator of protein kinase G(PKG), deduced the density of ICa-L by 10.50%±1.11%. Applying resveratrol and 8Br - cGMP simultaneously decreased the ICa-L significantly by 87. 58% ± 3.49% ( n = 6, P 〈 0. 05 ). ( 3 ) 5 μmol/L H8, a PKG inhibitor, inhibited the decrease in ICa-L caused by resveratrol. CONCLUSION: Resveratrol inhibits ICa-L in guinea pig ventricular myocytes, and this inhibitory effect involves the PKG pathway.
