摘要
目的观察叶下珠复方对刀豆蛋白A(ConA)所致免疫性肝损伤的保护作用。方法48只NIH小鼠随机分为6组,分别为空白对照组,模型对照组,叶下珠复方大、中、小剂量组,联苯双酯组(150mg/kg);除空白对照组外,所有小鼠于实验首日尾静脉注射ConA20mg/kg后,叶下珠复方大中小剂量组和联苯双酯治疗组均灌胃口服,空白对照组和模型对照组给予等容积生理盐水,每天一次,连续3d,末次给药后4小时,再次尾静脉注射ConA 20mg/kg,8h后取血检测ALT活性、TNF-α和IL-8含量,观察肝组织病理学变化。结果叶下珠复方大中小剂量组均能降低ALT、AST和TNF-α、IL-8水平,与模型对照组有显著性意义(均P<0.01),并在不同程度改善肝组织病理变化。但与联苯双酯组比较差异无显著性意义(P均>0.05)。结论叶下珠复方对ConA所致的小鼠免疫性肝损伤有保护作用。
Aim: to investigate the protective effects of Phyllanthus Urinaria L compound recipe against hepatic immune injury induced by concanavalinA (ConA) in mice .Methods:48 NIH mice were randomly divided 6 groups:normal control group(group A) ,model group(group B) ,high dosage group of Phyllanthus Urinaria L compound recipe ( groupC ), middle dosage group ( group D), low dosage group of Phyllanthus Urinaria L compound recipe (group E)and Bffendate group(group F) .Excepo the mice in group A,those in other groups were injected with ConA(2.0mg/kg) via the tail vain at the 1" day of experiment. The mice in Group C Group D and Group E were administered with Phyllanthus Urinaria L compound recipe at a dose 40g/kg.20g/kg and 10g/kg respectively. The mice in group E were administered with bifendate by gastric perfusion (150mg/kg). All the drugs were given once daily for 3 days consecutively. At the last time, when drugs were administered for 4 hours, the mice were injected with ConA once again at the same dosage. Blood sample were collected at the 8th hour after CortA administered for determining the activity of alanine transaminase (ALT) and aminotransferase (AST), the concentrations of IL-8 and tumor necrosis factor alpha (TNF-a). Histopathological examination for liver tissue was also performed. Results: compared with the model group, Phyllanthus Urinaria L compound recipe in three dosages markedly depressed serum levels of ALT and TNF-a,IL-8 expression( P 〈 0.01), and improved the hepatic pathological features. Conclusions: Phyllanthus Urinaria L compound recipe has a good protective effects against hepatic immune injury induced by CortA.
