臂丛损伤后细胞外三磷腺苷对脊髓组织中NT-3和NF-200表达的影响

The effect of exogenous ATP on expression of neurotrophin-3 and neurofilament protein in spinal cord after brachial plexus injury

摘要 目的观察腹腔注射细胞外三磷腺苷(ATP)对臂丛神经切断后脊髓组织中神经营养因子-3(neurotrophin-3,NT-3)和神经微丝蛋白(neurofilament protein,NFP)表达的影响。方法雌性Wister大鼠60只,体重280~300 g,随机分成2组,造成大鼠右侧臂丛神经根性切断损伤模型。手术后实验组每日腹腔注射三磷腺苷0.4 ml(2mg/kg),对照组腹腔注射生理盐水0.4 ml。分别于手术后2、4、6周取脊髓C_5~T_1组织,应用免疫组化染色和图象分析系统方法观察NT-3和NF-200表达。结果腹腔注射细胞外三磷腺苷能显著地增加臂丛切断损伤后脊髓组织中NT-3和NF-200的的表达(P<0.05)。结论腹腔注射细胞外三磷腺苷能增加臂丛损伤后脊髓组织的NT-3和NF-200的表达。 Objective To investigate the protective effect of extracellular ATP on spinal cord motor neurons in rats after brachial plexus root rhizotomy. Methods A total of 60 adult female Wistar rats weighing 280- 300 g were randomly divided into ATP and control groups. The right C5 - T1 nerve roots were transected. 0.4 ml (2 ng/kg·d) of ATP or normal saline respectively was given via intraperitoneal injection immediately and once daily to the rats. The rots were sacrificed on postoperative days 14, 28 and 42 respectively. The C6 - T1 segments of the spinal cord were harvested. Expression of neurotrophin-3 (NT-3) and the content of neurofilament protein (NFP) were determined by immunohistochemical technique and image analysis. Both NT-3 and NF-200 grey scale of motor neurons of the two groups were detected and compared. Results Compared to the control group, the expression of NT-3 and NF-200 was increased in the treated group with ATP injection ( P 〈 0.05). Cocnlusion Intraperitoneal injection of ATP could enhance the expression of NT-3 and NF-200 in tissues of the spinal cord.
