摘要
目的观察急性心肌梗死(acute myocardial infarction,AMI)患者外周血来源内皮祖细胞(endothelial progentior cells,EPCs)CXCR4受体表达及功能的变化。方法选择AMI患者15例,对照组非AMI患者15例,抽取外周血密度梯度离心法获取单个核细胞,培养7d后对贴壁的细胞进行分析。激光共聚焦显微镜鉴定FITC标记荆豆凝集素Ⅰ和DiI标记的乙酰化低密度脂蛋白双染色阳性细胞为EPCs。应用改良的Boyden小室测定EPCs对基质细胞衍生因子-1(stromal cell-derived factor-1,SDF-1)的迁移能力,流式细胞仪测定EPCs的CXCR4的表达,RT-PCR法测CXCR4的mRNA表达。结果AMI患者外周血EPCs数目增多,对SDF-1的迁移能力增强,其表达CXCR4上调,CXCR4的mRNA合成增强。与对照组相比,差异均具有统计学意义(P〈0.01)。结论AMI患者外周血来源EPCs的CXCR4受体表达上调,功能增强。
Objective To investigate the changes of expression and function of CXCR4 dwelling on the membrane of endothelial progentior cells (EPCs). Method Mononuclear cells were isolated from blood of patients with acute myocardial infarction (AMI, n = 15) and age-matched control subjects (n = 15) by density gradient centrifugation. Attaching cells were harvested after 7 days culture. EPCs were identified as adherent cells with double positive DiI-acLDL-uptake and lectin binding by direct fluorescent staining under a laser scanning cunfocal microscope. EPCs migration in response to stromal cell-derived factor-1 (SDF-1) was measured by a modified Boyden chamber assay. The expression of CXCR4 was analyzed by flow cytometry and mRNA was examined by RT-PCR. Results The number of EPCs was significantly increased in subjects with AMI. In addition, EPCs migration in response to SDF-1 was enhanced. The expression of protein and mRNA were also found up-regulated in EPCs. Conclusions This study demonstrates the up-regulated expression of CXCR4 on the EPCs from AMI and its enhanced function.
