摘要
目的:研究固本防喘胶囊对哮喘动物模型气道嗜酸性粒细胞凋亡的影响,为临床应用提供理论依据。方法:体重200～250g雌性豚鼠50只,随机分为正常对照组、模型组、西药对照组、中药小剂量治疗组、中药大剂量治疗组,每组10只。正常对照组以生理盐水进行腹腔注射和雾化。其余4组采用卵蛋白诱导法建立哮喘模型;在第一次诱喘24h后,正常对照组及模型组予生理盐水,西药组予地塞米松片(1mg.kg-1)胃饲;中药小剂量组及中药大剂量组分别予固本防喘胶囊浓缩液(每mL含生药2g)小剂量(2.5g.kg-1)及大剂量(7.5g.kg-1)胃饲治疗10天。疗程结束后,处死动物,收集各组动物肺组织标本,TUNEL法检测肺组织中气道嗜酸性粒细胞凋亡情况。结果:中药大、小剂量组豚鼠气道嗜酸性细胞凋亡明显增加,与哮喘模型组比较有显著性差异(P<0.01),大剂量组与西药对照组相当(P>0.05)。结论:固本防喘胶囊能诱导或促进哮喘豚鼠气道EOS凋亡,从而减轻气道炎性细胞浸润,控制气道慢性炎症,达到防治哮喘的作用。
Objective.To study the effect of Guben Fangchuan Capsule （固本防喘胶囊,GFC） on apoptosis of eosinophilic cells in guinea pigs. Methods ：Fifty cases of female GP （weight 200-250 g）were randomly divided into the normal group,the model group,the control group with WM,the treatment group with small TCM dosage ,the treatment group with big TCM dosage, Every group contains ten guinea pigs. The normal group were treated with intrapenitoneal injection and atomization with normal saline. The model group were treated with dexamethasone. The TCM groups with small and big dosages were gavaged with GFC in 2.5 g/kg or 7.5 g/kg. Apoptosis of eosinophilic cells in the lung tissue of guinea pigs were determined with TUNEL. Result：Apoptosis of eosinophilic cells in the TCM group with big dosage was increaed significantly compared with the model group （P 〈 0. 01）. Conclusion：Apoptosis of eosinophilic cells in guinea pigs were induced and enhanced by GFC,so the chronic inflammation in the airway was improved and the asthma was prevented.
