摘要
[目的]测定中药玫瑰花和月季花中槲皮苷的含量。[方法]采用反相高效液相色谱法,以Hypersil GOLD(5μm250×4.60mm)分析柱,乙晴:水(加磷酸调pH至2.6)(玫瑰花为20:80,月季花为16:84)为流动相,流速:0·8ml·min^-1;检测波长为254nm,柱温30℃。[结果]槲皮苷浓度在(0.005-0.200)μg范围内与色谱峰面积呈良好的线性关系,r=0.9998。[结论]该方法简易准确,重现性好,在本色谱条件及样品处理方法下,可以准确测出玫瑰花和月季花中槲皮苷的含量。
[Objective] To establish HPLC method for determination of the contents of quercitrin in Flos rosae rugosae and Flos rosae chinensis. [Method] The HPLC was conducted on a Hypersil GOLD column (5μm,250 min×4.6 mm) with Acetonitrilewater solution (20 : 80 for Flos rosae rugosae,16 : 84 for Flos rosae chinensis). The solution was adjusted to pH 2.6 with phosphoric acid and used as mobile phase with the flow rate of 0.8 mL·min^-1. The detective wavelength was set to 254 nm and column temperature at 30℃. [Result]The area under the peak of chromatography is very well correlated (r = 0. 9998) with the concentration of quercitrin in the range of (0. 005 -0. 200) μg. [Conclusion]This method is simple and accurate. The experimental results are very reproducible. Under the conditions as stated in the method section, the quercitrin can be separated very well from the Flos rosae rugosae and the Flos rosae Chinensis. The concentration of the quercitrin, therefore, can be accurately determined.
