黑曲霉固态发酵产酸性磷酸酯酶的工艺研究 被引量:1

摘要 为探讨黑曲霉固态发酵酸性磷酸酯酶(ACP)的产酶条件,首次采用固态培养的方法,在对黑曲霉3350产酸性磷酸酯酶的培养基组成和培养条件单因素研究的基础上,设计了Plackett-Burman试验选取了3个主要影响因素,并进行响应面分析。结果表明,黑曲霉3350产酸性磷酸酯酶的最适培养基和发酵条件为:麸皮8.0g,蔗糖2.0g,豆饼粉1.11g,水13.9ml,土温-801.0mL/kg,NH4NO3 20g/kg,KH2PO4 2.5g/kg,自然pH值;32℃固体静置培养120h,期间翻曲2~3次。在该条件下菌株最高产酶活性为154IU/g干曲,比优化前产酶提高88%,与液态发酵周期(9d)相比缩短了44%。 The research on the nutrition ingredients and fermentation conditions on the produetion of acid phosphatase by Aspergillus niger 3350 showed that the optimal fermentation medium and condition contained 8.0 g wheat bran, 2.0 g sucrose, 1 .11g soybean flour, 13.9 ml 1420, soil temperature-80 of 1.0 ml/kg, 20 g/kg, KH2PO4 2.5 g/kg, CaCl2 1.0 g/kg, MgSO4 1.0 g/kg, nature pH, solid culture for 20 h at 32℃, turning over the aspergillns 2 times. Under these conditions, the highest acid phosphatase activity was 154 IU/g dry matter, being 88% higher than acid phosphatase production before optimization.
