摘要 目的:造血干/祖细胞能够顺利的归巢至骨髓造血微环境是干细胞移植成功的关键因素之一。在复杂的归巢过程中,造血干/祖细胞与骨髓微环境中的各种成分及黏附分子相互作用维护造血平衡。本文就黏附分子的种类、结构、功能及其在造血干/祖细胞归巢中的作用进行文献综述。资料来源:应用计算机检索Pubmed和Springer数据库2000-01/2006-11期间的相关文章,检索词为"hematopoieticstem/progenitor cell,adhesion molecule,homing",并限定文章语言种类为English。同时计算机检索CNKI中国全文期刊数据库2000-01/2006-11期间的相关文章,检索词为"造血干/祖细胞,黏附分子,归巢",并限定文章语言种类为中文。资料选择:对资料进行初审,并查看每篇文献后的引文。纳入标准:①文章所述内容应涉及黏附分子的种类、结构、功能及其在造血干/祖细胞归巢中的作用。②2000年以后发表的文章。排除标准:重复研究或Meta分析类文章。资料提炼:共收集到42篇相关文献,30篇文献符合纳入标准,排除的12篇文献为内容陈旧或重复。符合纳入标准的30篇文献中,分别涉及黏附分子的种类和结构、黏附分子在造血干细胞归巢中的作用等内容。资料综合:造血干细胞移植是治疗恶性血液病和多种实体肿瘤的有效方法,造血干细胞顺利归巢至骨髓是造血干细胞移植成功的关键因素之一,在归巢过程中黏附分子起到十分重要的作用。近年来有关造血干细胞归巢及黏附分子的医学研究进展,具体内容可被归纳为以下两个方面:①黏附分子的种类、结构及功能。②黏附分子在造血干细胞归巢过程中的作用。结论:黏附分子与造血干细胞归巢有着十分密切的关系,但究竟是哪些黏附分子在归巢中发挥决定性作用尚无一致性结论。随着对造血干细胞归巢机制的深入研究,将为造血干细胞移植在临床的广泛应用带来更加广阔的前景。 OBJECTIVE: One of the key factors in successful stem cell transplantation is the hematopoietic stern/progenitor cell could home to bone marrow microenvironment. In the complicated process, hematopoietic stem/progenitor cells, components of the bone marrow microenvironment and adhesion molecule interact with each other to keep hematopoietic balance. In this review, the variety, structure and function of adhesion molecule and its role in the homing of hematopoietic stem/progenitor cell are summarized. DATA SOURCES: A computer-based online search of Pubmed, Springer database was undertaken to identify the relevant articles published in English between January 2000 and November 2006 with the key words of "hematopoietic stem/progenitor cell, adhesion molecule, homing'. Meanwhile, we searched the CNKI database for related articles published in Chinese from February 2000 to November 2006 with same key words in Chinese. STUDY SELECTION: All articles were selected firstly, and the references of each article were looked over. Inclusive criteria: ①Articles on the variety, structure and function of adhesion molecule and its role in the homing of hematopoietic stem/progenitor cells; ②Articles published after 2000. Exclusive criteria: Repeated study and Meta analysis papers. DATA EXTRACTION: A total of 42 relevant literatures were collected, and 30 were in accordance with the inclusive criteria; 12 old and repetitive articles were excluded. The 30 enrolled literatures referred to the variety, structure and function of adhesion molecule and its role in the homing of hematopoietic stem/progenitor cell. DATA SYNTHESIS:Hematopoietic stem cells transplantation is an effective treatment for malignant hematopathy and various solid tumors, and the successful homing of hematopoietic stem cells to bone marrow is one of the critical factors in the transplantation. Adhesion molecule plays an important role in the homing process. The recent development of hematopoietic stem cells homing and adhesion molecule could be listed as follows:①The variety, structure and function of adhesion molecule; ②the role of adhesion molecule in the homing process of hematopoietic stem cells. CONCLUSION: Adhesion molecules are closely related to the process of hematopoietic stem cells homing, however, which kinds of molecule play the decisive function in this process is not a uniform conclusion. With the further research on the mechanism of hematopoietic stem cells homing, it will offer a wide prospect for the application of hematopoietic stem cells transplantation in clinic.
