摘要
以美国黑树莓叶片作为外植体,以MS为基本培养基,添加不同浓度的6-BA、NAA或IAA进行愈伤组织诱导及植株再生试验。结果表明:树莓叶片外植体在培养基MS+6-BA 2.0-4.0 mg/L+NAA(IAA)0.01-0.5 mg/L都能不同程度地形成愈伤组织,但以MS+6-BA4.0 mg/L+NAA 0.5 mg/L和MS+6-BA 4.0 mg/L+IAA 0.5 mg/L愈伤组织的诱导率最高,诱导率可达100%;所试验的各种不定芽分化培养基,都能不同程度地诱导愈伤组织分化出不定芽,在添加6-BA 2.0 mg/L和NAA 0.01 mg/L的MS培养基上,愈伤组织分化不定芽的比例最高,达49.7%的;所产生的不定芽在1/2 MS+NAA 0.1 mg/L的培养基上的生根率可达89.3%。
The leaves of the blackberry as the explant,and MS contained different concentration of 6-BA, NAA or IAA as the media, the callus induction and the plant regenention of the blackberry were studied. The results showed that the calluses were induced from the leaf explants on the media of MS + 6-BA 2.0- 4.0 mg/L + NAA 0.01 -0.5 mg/L and MS + 6-BA 2.0- 4.0 mg/L + IAA 0.01 -0.5 mg/L. The induction rate was up to 100% on the media of MS + 6-BA 4.0 mg/L + NAA 0.5 mg/L and MS + 6-BA 4.0 mg + LAA 0.5 mg/L. Adventitious buds were induced from the calluses at the frequency of 49.7% on MS medium supplemented with 6-BA 2.0 mg/L and NAA 0.01mg/L. 89.3% adventitious buds were rooted and irduced regenerated plants on 1/2 MS medium supplemented with NAA 0.1 mg/L.
