白细胞介素-10对人牙囊细胞OPG表达的影响 被引量:4

Regulation of OPG secretion by IL-10 in human dental follicle cells.

摘要 目的:研究人牙囊细胞白细胞介素-10(IL-10)蛋白的表达及其对骨保护素(osteoprotegerin,OPG)表达的影响。方法:采用组织块加胶原酶消化法培养人牙囊细胞,进行IL-10免疫组化染色。将25ng/mlIL-10作用于牙囊细胞0h、1h、3h、6h、9h,用ELISA法检测上清液中OPG含量变化。结果:人牙囊细胞IL-10表达阳性。25ng/mlIL-10作用于人牙囊细胞3~6hOPG表达显著增加(p<0.05)。结论:人牙囊细胞表达IL-10,IL-10通过增加人牙囊细胞OPG表达,在牙齿萌出过程中起重要的调控作用。 Objective:To study the expression of IL-10 at protein level in human dental follicle cells (HDFCs)and the effect of IL-10 on osteoprotegerin (OPG) secretion. Method: HDFCs were cultured by explant method. The expression of IL-10 at protein level was tested by immunocytochemistry and OPG secretion treated with 25 ng/ml IL-10 at 0, 1,3,6,9 h was examined by ELISA method. Result: IL-10 was expressed in the cytoplasm of HDFCs. OPG secretion was up-regulated by IL-10 at 3-6h(p〈0.05). Conclusion:IL-10 may regulate tooth eruption by up-regulating OPG secretion.
