摘要
首次尝试在培养细胞的化学转化实验中应用整装细胞内质网技术以观察细胞超微结构及分布变化与细胞表型的关系,实验结果证实整装细胞内质网方法可清晰显示转化中细胞的超微结构,分布及相邻细胞器的全貌,并观察到处理组内质网分布变化同期伴有细胞表型改变,表现为高饱和生长密度,接触抑制缺失,细胞多型性等,这种一致性证明内质网与细胞癌变密切相关,整装内质网技术是一项较好而实用的检测早期转化细胞超微结构改变的细胞学方法,该实验为今后探讨转化中细胞超微结构整体变化提供了新的研究手段及模型系统,并从整体的角度丰富了转化细胞超微结构的科学资料。
The present experiment appears to be the first visual demonstration on relation between the distribution of endoplasmic reticulum(ER) in wholemount cultured cell and cell phenotype in vitro neuoplastic transformation by chemical carcinogen. New ER preparation technique has been used to study ER architedchques of cell in early chemical carcinogenesis. The sesult showes well defined ER ultrustrucure and distribution as well as neibouring organells in the cells. Significant difference of ER distribution between treated and control group at different periods could be observed. Some phenotype alteration related to early transformation with ER disrtribution alteration in treated group is also noticed. The technique is a new model system for detecting cytologic signs in early carcinogenesis and the result suggests ER is closely related to carcinogenesis.
